The Effects of Macrophages on the Wound Healing Delayed by Immunosuppression


Abstract
The effects of macrophages on the wound healing delayed by immunosuppression were studied in Balb/C or ICR mice. The immunosuppression were induced by ultraviolet radiation (UVR), lipopolysaccharide(LPS) or glcocorticod (GCS) treatment. After the
immunosuppression, the full thickness wounds were made by 5mm paper puncher on the backs of the mice. The peritoneal macrophages from the syngeneic mice or RAW 264.7 cells were injected around the wound. The wound sizes and histological findings
were
observed periodically.
@ES The results were as follows:
@EN 1) The number of leukocytes and monocytes of the peripheral blood were not changed for 3 days despite of the local injections of macrophages in both normal and immunosuppressed mice.
2) The wound sizes of the normal mice were not changed by local injection of peritoneal macrophages.
3) The times for the recovery of the wounds in the immunosuppressed mice induced by UVR, LFS or GCS were shortened by the local injections of peritoneal macrophages (1¡¿10E5/wound).
4) The time for the recovery of the delayed wound healing was shortened in the group of macrophage injection just after of the wound formation than that of macrophage injection 24 hour after the wound formation in the immunosuppressed mice.
5) Recovery of the dealyed wound healing was induced by local injection of RAW 264.7 cells. These were more effective in the group of activated RAW 264.7 cells by IFN-¥ã.
6) The mononuclear cells were reduced in number around the wound in the immunosupressed mice through the experimental periods and these findings were similar I the macrophage injection groups. The injected macrophages were destructed in 3 days
after
injection.
The results of this study suggest that the local injection of macrophages around the wound can shorten the time for the wound healing delayed by immunosuppression and these effects might be induced by the secretory products or contained certain
materials in the macrophages.
